Sinner defeated 26th-ranked player Tallon Griekspoor of the Netherlands 5:7, 7:5, 6:1 and will face 66th-ranked player Christopher O'Connell of Australia in the next match.
Daniil Medvedev defeated Cameron Norrie, ranked 31st in the world player rankings, 7:5, 6:1 in the third round.
Medvedev's next opponent will be the world's 50th-ranked player, Germany's Dominik Köpfer.
The prize fund of the tournament in Miami is $8.9 million.
